Properties of Polyphenol Oxidase from Artemisia princeps var. orientalis Leaves

Abstract
PPO was purified from the extract of Artemisia princeps var. orientalis leaves by ammonium sulfate fractionation and sephadex G-150 column gel filtration chromatography, which resulted in a 90-fold increase relative to homogenate without inhibitors in specific activity. The optimum temperature was 25oC and the optimum pH was 7.0 in enzymic reaction. The best substrates were catechol and 4-methylcatechol with Km values of 3.26mM, 1.03mM, respectively. The enzyme possesses high catecholase activity but little cresolase activity. The enzyme was inhibited in the presence of sod. diethyldithiocarbamate, KCN, thiourea and EDTA. Among the divalent cations(Cu2+, Zn2+, Mn2+, Mg2+, Co2+, Ni2+), Cu2+ and Zn2+ were activators on PPO, whereas Mg2+ and Co2+ were inhibitors on PPO.
